To establish a method to produce highly pure [125I] 19-nortestosterone derivatives suitable for biological investigations. Allopregnanolone- and progesterone-nortestosterone derivatives were synthesised by conjugating bovine serum albumin (BSA) with the 3-iodo-group of the hormone via the carboxyl function of the pyridine ring. The binding capacity of these conjugates to the guinea pig brain gamma-aminobutyric acid (GABA) receptor was determined and compared with that of [125I]GABA. Radioiodinated test conjugates were stored at 4 degrees C, at room temperature, and under physiological conditions. Binding capacity decreased after 4 weeks at 4 degrees C. By increasing the temperature to room temperature the binding capacity was slightly affected. Storage of the radioiodinated conjugates under physiological conditions resulted in rapid deterioration of binding capacity. The results indicate that allopregnanolone-nortestosterone and progesterone-nortestosterone derivatives possess an unstable binding affinity towards the gamma-aminobutyric acid (GABA) receptor and should be stored at low temperatures (4 degrees C).
